Experts from Durham University have identified a new gene that could help the development of fertility treatments in humans in the future.

Scientists from Durham University, UK, and Osaka University, Japan, looking at fertility in mice, have discovered for the first time that the gene, which makes a protein called PDILT, enables sperm to bind to an egg, a process essential to fertilisation.

The team found that when the gene was 'switched off' in male mice, less than three per cent of females' eggs were fertilised compared to more than 80 per cent in mice when the gene was left switched on.

It is the first time that a gene of this type has been linked to fertility.

The researchers also found that the cumulus cells, a cluster of cells surrounding and protecting an egg, play an important role in fertility - their presence enables sperm to bind properly to an egg.

Although the research and findings are at an early stage, the researchers now hope to look at how the gene affects sperm-to-egg binding in humans. The findings are published in the journal PNAS, Proceedings of the National Academy of Sciences.

The discoveries could pave the way for the development of new fertility treatments for humans that could help to reduce the cost and improve the effectiveness of in vitro fertilisation (IVF). They could also assist research into new contraceptive techniques that deactivate the gene and prevent sperm reaching an egg and binding to it.

Dr Adam Benham, School of Biological and Biomedical Sciences, Durham University, said: "The protein is an essential part of the navigation system of sperm. It helps sperm swim through the oviduct to the egg and without it sperm get stuck. Our results show that navigating the oviduct is an important part of the fertilisation process.

"Like any navigation system, you have to programme where it is that you want to go and this protein plays an essential role in getting sperm to the right destination, in good shape, and in good time."

The researchers found the gene a few years ago through a database search for new genes of the PDI family. Following extensive research, they established that the gene made an important protein in the testes.

The PDILT gene, part of the PDI family, helps another gene product called ADAM3 to form and assemble correctly, and then to reach the surface of a sperm, thereby equipping it with the right tools and machinery to navigate and make contact with an egg.

The new findings show the importance of PDILT in the process of sperm-to-egg binding and in enabling sperm to swim past the uterus, ascend the oviduct and to get through the sticky outer layers of an egg.

The team honed in on the role of the protein by switching it off in mice and tracking the ability of sperm to bind to and fertilise eggs in Petri dishes and in mice. They noticed that sperm from mice with the gene switched off will not bind to a bare egg, but will bind to an egg surrounded by cumulous cells.

Dr Benham said: "We now hope to discover how the PDILT protein affects fertility in humans. Mutations in the gene may be responsible for unexplained male fertility problems and further research may aid more effective IVF treatment."

Dr Masahito Ikawa from Osaka University, Japan, added: "This protein is essential for sperm to migrate successfully and is required for fertility. The next step is to see how this protein works with other proteins to control the sperm binding and fusion process."

Israeli prime minister explores early elections

JERUSALEM (AP) ? In a political development with global implications, Prime Minister Benjamin Netanyahu on Sunday signaled he soon might call early elections ? a decision that could put Mideast peace efforts on hold for months and cast more uncertainty on Israel's deliberations over whether to attack Iran's nuclear program.

Netanyahu has presided over an awkwardly diverse yet unusually resilient coalition for three years and is suddenly beset by challenges. But while he is increasingly under fire for his handling of Iran and the Palestinians, it is more mundane domestic complications that appear to be driving the coalition unrest.

The Supreme Court has ordered the government to dismantle illegal West Bank settlement outposts ? a move that faces tough opposition from hard-line coalition allies who have a history of toppling even right-leaning governments like Netanyahu's over such issues.

A Supreme Court-ordered Aug. 1 deadline to scrap draft exemptions for tens of thousands of religious seminary students ? an issue dividing the country's Jews into increasingly antagonistic camps ? also threatens the coalition.

Mass protests also are expected for the second summer in a row over Israel's high cost of living.

If balloting is held soon, Netanyahu's chances of re-election seem good ? a remarkable situation given the sour national mood, the threat of regional war, and the forces arrayed against him. Israel's own elites and the world community seem largely united in preferring his demise.

The government's term can last through October 2013 ? but Israeli governments rarely serve a full term.

An Israeli official said Netanyahu would consult with coalition partners in the next week before deciding on a new date.

The official said Netanyahu's main concern was that his partners would make excessive budget demands for 2013 and quoted him as saying he wouldn't cave in to "extortion." The official spoke on condition of anonymity because he was discussing a closed meeting.

Netanyahu also told leading members of his Likud Party that he was weighing the possibility, and early elections were the focus of media reports and talk shows all day. Israel's respected Channel 10 TV cited unidentified official sources as saying elections would take place within six months.

The opposition "will be ready for elections in October," said Shaul Mofaz, a former military chief recently elected to head Kadima, the largest opposition party.

With the budget season months away, Netanyahu faces more immediate threats. The Supreme Court has ordered the government to dismantle an illegally built West Bank settlement outpost by Tuesday. Pro-settlement lawmakers have warned an evacuation of the Ulpana outpost could bring down the coalition. Netanyahu has asked the court for a delay.

Even if he weathers that crisis, he faces a far larger battle over the religious draft exemptions. The exemptions have long enraged Israel's secular majority, whose young men and women serve years of mandatory military service while ultra-Orthodox men participate instead in government-subsidized religious studies.

Many continue studies for years, leading to expanding ultra-Orthodox dependence on welfare. The animosities grow daily, with increasing friction over ultra-Orthodox demands such as gender segregation in some situations.

Netanyahu's two largest coalition partners, the ultra-Orthodox Shas party and the secular Yisrael Beitenu group, are deeply at odds over the exemptions. Losing either party would rob him of his parliamentary majority.

Shas officials said Sunday they are not afraid of new elections over the issue. "It is delusional to think you can impose such a drastic and substantial social maneuver on an entire segment of the population," said Nissim Zeev, a Shas lawmaker.

Yisrael Beitenu's leader, Foreign Minister Avigdor Lieberman, threatened this weekend to bring down the government if a compromise isn't found. He said he would submit on May 9 a proposal for no more than 1,000 exemplary religious scholars to be granted exemptions. "Our commitment to the coalition has ended. Now we have a commitment to the voter," Lieberman said Sunday.

Netanyahu met Sunday with a group of army reservists and promised the existing law would be replaced by a more "just and egalitarian law that would more fairly divide the burden among all Israeli citizens."

In another complication, it is expected that new legislation would also seek to impose civilian national service on Israel's 1.5 million Arab citizens. That would almost surely antagonize the Arab sector on which Israel's dovish opposition depends for any chance of a majority. Their current exemption is a double-edged sword, reflecting de facto segregation and a state reluctant to test their loyalty.

A poll published Sunday indicated Netanyahu remains on course for re-election ? predicting his Likud party would win 31 seats, up from its current 27, making it the largest faction in the 120-seat parliament. Parties supporting him would retain their overall majority. The poll, conducted last week by the New Wave Research Polling Institute, questioned 500 people and had a margin of error of 4.5 percentage points.

An election could yield a different Israel even if Netanyahu wins, however, since it is widely believed that Mofaz and other opposition figures would join him in a centrist coalition, freeing him of the grip of nationalist and religious parties.

An extended election campaign would cast new uncertainty on the country's two most pressing foreign policy matters: peacemaking with the Palestinians and Israel's claims that time is running out for military action against Iran's suspected nuclear weapons program.

Netanyahu has come under heavy criticism for his handling of both issues ? most recently over the weekend by Yuval Diskin, the recently retired head of the Shin Bet internal security service.

Diskin became the latest in a series of figures from the security establishment to express concern about Netanyahu, and similar sentiments are often voiced in academic, business and judicial circles.

"All the heads of the security establishment, including current ones, oppose (Netanyahu's) policy," said Channel 10 analyst Emanuel Rosen, predicting the issue could affect the election campaign in a country that still lionizes military figures.

In comments posted on YouTube, Diskin said he had "no faith" in Israel's leadership. He accused the government of exaggerating the effectiveness of a possible military attack on Iran and not actively pursuing peace with the Palestinians.

Netanyahu is unlikely to offer any bold concessions in an election campaign. With the U.S., the main Mideast mediator, focused on its own election, continued deadlock appears likely through the end of the year.

It remains unclear what effect elections would have on the Iran issue. It is unclear how much damage Israel could do to Iran's fortified nuclear facilities, and such an attack would invite retaliation and anger from the U.S. and other key allies who are seeking a diplomatic solution with Tehran.

On the other hand, a surprise attack could give Netanyahu an electoral boost. In 1981, then-Prime Minister Menachem Begin ordered a surprise strike that destroyed an Iraqi nuclear reactor just three weeks before elections. Begin's opponents accused him of political motivations, and he was re-elected by a whisker.

While chlorpyrifos is no longer registered for household use in the US, it continues to be widely used around the world, as well as on many food and agricultural products throughout the US

Even low to moderate levels of exposure to the insecticide chlorpyrifos during pregnancy may lead to long-term, potentially irreversible changes in the brain structure of the child, according to a new brain imaging study by researchers from the Columbia Center for Children's Environmental Health at the Mailman School of Public Health, Duke University Medical Center, Emory University, and the New York State Psychiatric Institute. The changes in brain structure are consistent with cognitive deficits found in children exposed to this chemical.

Results of the study appear online in the April 30 PNAS.

The new study is the first to use MRI to identify the structural evidence for these cognitive deficits in humans, confirming earlier findings in animals. Changes were visible across the surface of the brain, with abnormal enlargement of some areas and thinning in others. The disturbances in brain structure are consistent with the IQ deficits previously reported in the children with high exposure levels of chlorpyrifos, or CPF, suggesting a link between prenatal exposure to CPF and deficits in IQ and working memory at age 7.

The study also reports evidence that CPF may eliminate or reverse the male-female differences that are ordinarily present in the brain. Further study is needed to determine the consequences of these changes before and after puberty, the researchers say.

Notably, the brain abnormalities appeared to occur at exposure levels below the current EPA threshold for toxicity, which is based on exposures high enough to inhibit the action of the key neurological enzyme cholinesterase. The present findings suggest that the mechanism underlying structural changes in the brain may involve other pathways.

According to the lead author, Virginia Rauh, ScD, Professor at the Mailman School of Public Health and Deputy Director of the Columbia Center for Children's Environmental Health, "By measuring a biomarker of CPF exposure during pregnancy, and following the children prospectively from birth into middle childhood, the present study provides evidence that the prenatal period is a vulnerable time for the developing child, and that toxic exposure during this critical period can have far-reaching effects on brain development and behavioral functioning."

"By combining brain imaging and community-based research, we now have much stronger evidence linking exposure to chlorpyrifos with neurodevelopmental problems," adds senior author Bradley S. Peterson, MD, Chief of Child & Adolescent Psychiatry, New York State Psychiatric Institute, and Director of MRI Research in the Department of Psychiatry, Columbia University Medical Center.

In the current study, the researchers used MRI to evaluate the brains of 40 New York City children, ages 5 to 11, whose mothers were enrolled prenatally in a larger cohort study. Researchers compared 20 children with high exposures to CPF with 20 children with lower exposures; all exposures occurred prior to the EPA ban on household use of the chemical in 2001. They found brain anomalies were associated with the higher exposure.

While chlorpyrifos is no longer registered for household use in the US, it continues to be widely used around the world, as well as on many food and agricultural products throughout the US

Even low to moderate levels of exposure to the insecticide chlorpyrifos during pregnancy may lead to long-term, potentially irreversible changes in the brain structure of the child, according to a new brain imaging study by researchers from the Columbia Center for Children's Environmental Health at the Mailman School of Public Health, Duke University Medical Center, Emory University, and the New York State Psychiatric Institute. The changes in brain structure are consistent with cognitive deficits found in children exposed to this chemical.

Results of the study appear online in the April 30 PNAS.

The new study is the first to use MRI to identify the structural evidence for these cognitive deficits in humans, confirming earlier findings in animals. Changes were visible across the surface of the brain, with abnormal enlargement of some areas and thinning in others. The disturbances in brain structure are consistent with the IQ deficits previously reported in the children with high exposure levels of chlorpyrifos, or CPF, suggesting a link between prenatal exposure to CPF and deficits in IQ and working memory at age 7.

The study also reports evidence that CPF may eliminate or reverse the male-female differences that are ordinarily present in the brain. Further study is needed to determine the consequences of these changes before and after puberty, the researchers say.

Notably, the brain abnormalities appeared to occur at exposure levels below the current EPA threshold for toxicity, which is based on exposures high enough to inhibit the action of the key neurological enzyme cholinesterase. The present findings suggest that the mechanism underlying structural changes in the brain may involve other pathways.

According to the lead author, Virginia Rauh, ScD, Professor at the Mailman School of Public Health and Deputy Director of the Columbia Center for Children's Environmental Health, "By measuring a biomarker of CPF exposure during pregnancy, and following the children prospectively from birth into middle childhood, the present study provides evidence that the prenatal period is a vulnerable time for the developing child, and that toxic exposure during this critical period can have far-reaching effects on brain development and behavioral functioning."

"By combining brain imaging and community-based research, we now have much stronger evidence linking exposure to chlorpyrifos with neurodevelopmental problems," adds senior author Bradley S. Peterson, MD, Chief of Child & Adolescent Psychiatry, New York State Psychiatric Institute, and Director of MRI Research in the Department of Psychiatry, Columbia University Medical Center.

In the current study, the researchers used MRI to evaluate the brains of 40 New York City children, ages 5 to 11, whose mothers were enrolled prenatally in a larger cohort study. Researchers compared 20 children with high exposures to CPF with 20 children with lower exposures; all exposures occurred prior to the EPA ban on household use of the chemical in 2001. They found brain anomalies were associated with the higher exposure.

Kenya official: Grenade kills 1 in church attack

(AP) ? One person died and 15 people were wounded when a grenade was thrown into a church in Kenya's capital during Sunday service, an official said.

Nairobi's deputy police chief, Moses Ombati, said the grenade exploded at God's House of Miracles International Church in Nairobi.

The incident is the latest in a string of grenade attacks since Kenya sent troops into Somalia in October last year. Al-Qaida-linked al-Shabab militants from neighboring Somalia have vowed to carry out a major attack on Kenya for sending troops in.

Sunday's attack follows a U.S. Embassy warning that a terror attack on prominent government buildings and hotels in Nairobi could be imminent.

The embassy said Monday that the timing of the attack is not known but they believe it to be at a final planning stage.

Police have said that the Somali militant group is suspected of killing at least 30 Kenyan civilians since Kenyan troops entered Somalia.

In February, police spokesman Eric Kiraithe said the killings ? which go back to October ? are believed to have been carried out by sympathizers of al-Shabab in Kenya.

He said most of the attacks were carried out in towns near the border between Somalia and Kenya.

Police say that dozens of Kenyan youth have been recruited by al-Shabab and are operating in the country. Al-Qaida announced in February that it was merging with al-Shabab.

(AP) ? One person died Saturday and more than a dozen were taken to a hospital with injuries after high winds blew over a beer tent near Busch Stadium in St. Louis.

At least five others were critically injured and up to 100 people were treated at the scene after straight-line winds whipped through a large tent near Kilroy's Sports Bar, where about 200 people were celebrating after the Cardinals beat Milwaukee 7-3 earlier in the afternoon, officials said.

Eddie Roth, a spokesman for the St. Louis Department of Public Safety, said winds of about 50 mph shattered aluminum poles that held up the tent, which was located south of the stadium. The force of the wind blew the tent onto an adjacent railroad bridge.

"It was crazy, scary," said Annie Randall, whose family owns Kilroy's. "We're just so sorry this happened."

Two hours after the tent collapsed, tornado sirens were sounding across St. Louis as a severe thunderstorm capable of producing a tornado rolled through. Several people at Kilroy's were ushered into the basement of the business until the storm passed.

Bar owner Art Randall told the St. Louis Post-Dispatch things were flying through the air sideways and metal chairs were being flung across the beer garden when the storm hit.

Roth said the man who died appeared to be in his 50s, but he didn't know the extent of injuries sustained by other victims.

Roth also said the tent had passed inspection and it didn't appear there would be any violation, although the investigation is ongoing.

Fruit, veggies tied to lower diabetes risk

NEW YORK (Reuters Health) - People who get a range of fruits and vegetables in their diets may have a somewhat decreased risk of type 2 diabetes, a new study suggests.

The findings, reported in the journal Diabetes Care, do not prove that eating your fruits and veggies will ward off type 2 diabetes -- a disease closely associated with obesity and old age.

But researchers say the findings should give people yet more incentive to eat the way our mothers always told us.

The study of over 3,700 UK adults found that those who downed the most servings of fruit and vegetables in a week had a lower risk of developing type 2 diabetes over 11 years, versus people who ate the fewest.

Diabetes risk was also lower among people who ate a wider variety of fruit and vegetables -- regardless of the actual quantity they ate.

That suggests people should not only focus on how many servings they get each day, according to senior researcher Nita G. Forouhi, of the Institute of Metabolic Science in Cambridge, UK.

"The finding on variety of intake is new and exciting," she said in an email, "because it demonstrates that independent of the quantity consumed, we have the potential to gain additional and important benefits from choosing a mixture of different fruits and vegetables as part of a balanced diet."

For the study, Forouhi's team looked at data from 3,704 adults ages 40 to 79 that were part of a larger study on nutrition and chronic diseases.

Of those people, 653 developed type 2 diabetes over 11 years.

All of the study participants had kept a week-long food diary at the study's start. And Forouhi's team found that those who'd reported the highest combined fruit and vegetable intake were less likely to develop diabetes over the coming years.

Of the one-third with the highest intake -- typically about six servings of fruit and vegetables per day -- 16 percent were diagnosed with type 2 diabetes.

That compared with 21 percent of the one-third of participants with the lowest fruit and vegetable intake (about two servings per day).

That low-intake group closely matches the average American's diet. U.S. studies suggest that adults typically get two to three servings of fruit and vegetables combined each day.

Of course, fruit and veggie lovers may be different from non-lovers in a number of ways, Forouhi acknowledged -- including weight, exercise levels, smoking habits and education.

But when her team accounted for those factors, a high intake of fruit and vegetables was still linked to a 21 percent lower risk of developing type 2 diabetes.

The researchers found a similar pattern when it came to variety. People who averaged 16 different types of fruit and vegetables per week were about 40 percent less likely to develop diabetes than people who averaged eight types.

The findings do not prove that fruit and veggies deserve the credit. It's possible some other, unmeasured difference between study participants is at work, Forouhi said.

Still, the results underscore the standard diet advice for overall health: Eat your fruits and vegetables.

Variety may be key, Forouhi said, because that helps ensure you get a range of nutrients. That includes not only vitamins and minerals, but also fiber and plant compounds called phytochemicals, which are thought to help protect cells from damage that can lead to chronic disease.

People differ in how many servings of fruit and vegetables they need based on their weight or activity levels, for instance. But one common guideline is to go for four to five daily servings each of fruit and vegetables.

One serving is equal to a half-cup of cooked vegetables or a medium-sized piece of fresh fruit.

To get a good variety, Forouhi suggested incorporating a range of colors into your fruit-and-vegetable repertoire.

But, she added, that all needs to be part of a generally healthy lifestyle.

One of the biggest factors in type 2 diabetes risk is obesity. So experts generally advise maintaining a healthy weight by exercising regularly and watching calories.

If there is one thing we have learned in MTV's Battle of the Boy Bands, it's that all those boy band fans are extremely dedicated. Fans have proudly bonded together to show their allegiance to their favorite groups to help them advance to the next round, and as a result, we have had some shocking eliminations.

Yet, this bracket has also served as a reminder of why we fell in love with these groups to begin with. Whether it was their boyish good looks or catchy hit songs that hooked them, most people can probably name their favorite boy band of all time. So when we got the opportunity to sit down with Selena Gomez when she was promoting her new fragrance, we had to ask, which boy band stole her heart in her early teens?

"'NSYNC all the way, of course," Gomez revealed. "It was the first for me and then they did that tour with Britney [Spears] and I like lost my mind and will always remember that time in my life."

And when you have a favorite group, you have to have a favorite member, and Gomez's boy band crush coincidently has the same name as her current beau.

"It was Justin Timberlake," Gomez revealed. "But I think I also, I think I liked Lance Bass as well. ... He has pretty eyes."

Unfortunately for Gomez, 'NSYNC were knocked out in a battle against British boy band 5ive in the first round of competition. Luckily, she can still vote for the new wave of boy bands, The Wanted and One Direction.

"It's cool, that's kind of awesome that they're coming back and they legitimately are," Gomez said of boy bands making a comeback. "I like it."

China oil spill: CNOOC, ConocoPhillips agree to pay $266 million in compensation

China National Offshore Oil Corp (CNOOC) and ConocoPhillips have agreed to pay $1.68 billion yuan ($266 million) in compensation for damage caused by a series of oil spills off the coast of northern China last year, the Financial Times reported today.

CNOOC, China?s largest offshore oil producer, will also pay 480 million yuan to ?resume the social responsibility for Bohai environmental protection,? the company said in a statement.

The money will be used to fund environmental programs such as ?ecological and environmental protection? and restoration of the ?damaged marine ecological environment?.

Energy-hungry China is one of the world?s largest consumers of crude and is no stranger to major oil spills.

In 2010, an oil pipeline explosion in the northeastern city of Dalian sent oil across an area of 165 miles.

Teens with diabetes have trouble managing it, study says

New research sends a stark warning to overweight teens: If you develop diabetes, you'll have a very tough time controlling it.

A major study released Sunday tested several ways to manage blood sugar in teens newly diagnosed with diabetes and found that nearly half of the teens failed to control their blood sugar within a few years and that 1 in 5 suffered serious complications. The results spell trouble for a nation facing rising rates of "diabesity," Type 2 diabetes brought on by obesity.

The federally funded study is the largest look at how to treat diabetes in teens. Earlier studies have focused mostly on adults, and most diabetes drugs aren't even approved for youths. The message is clear: Prevention is everything.

"Don't get diabetes in the first place," saidDr. PhilZeitler of the University of Colorado, one of the study leaders.

A third of American children and teens are overweight or obese. They are at higher risk of developing Type 2 diabetes, in which the body can't make enough insulin or can't use what it does make to process sugar from food. Until the rise of the obesity epidemic, doctors rarely saw children with Type 2 diabetes. The more common kind of diabetes in children is Type 1, which used to be called juvenile diabetes.

Doctors usually start Type 2 treatment with metformin, a pill to lower blood sugar. If it still can't be controlled, other drugs and daily insulin shots may be needed. The longer blood sugar runs rampant, the greater the risk of vision loss, nerve damage, kidney failure, limb amputation, even heart attacks and strokes.

The goal of the study was simple: What's the best way for teens to keep diabetes in check?

The study involved 699 overweight and obese teens recently diagnosed with diabetes. All had their blood sugar normalized with metformin, then received one of three treatments to try to maintain that control: metformin alone, metformin plus diet and exercise counseling, or metformin plus a second drug, Avandia.

After nearly four years, half in the metformin group were not able to maintain blood sugar control. The odds were a little better for the group that took the two drugs but not much different for those in the lifestyle group.

Even so, Zeitler said, doctors would not recommend the combination drug therapy because Avandia has been linked to higher risk of heart attacks in adults. Those risks became known after this study began.

Another study leader from Children's Hospital Los Angeles, Dr. Mitchell Geffner, agreed that Avandia can't be recommended for teens, but said the study made clear they would need more than metformin to control their disease.

"A single pill or single approach is not going to get the job done," he said.

Among all the teens in the study, 1 in 5 had a serious complication such as very high blood sugar, usually landing them in the hospital.

The results were published online Sunday by the New England Journal of Medicine and presented at a pediatric meeting in Boston. The National Institutes of Health funded the study and drug companies donated the medications.

The Salzburg Connection. The city of Salzburg is indeliby associated with the name of Mozart, who lived and worked there for 25 years. But the city had a rich musical heritage going back over?100 years, particularly in?church music. One of Mozart's most illustrious predecessors was Heinrich Biber, active as a violinist and composer at the turn of the seventeenth century. Tonight's concert will feature two items of church music first performed in Salzburg Cathedral, Biber's Marian Vespers (1693) and Mozart's 'Coronation' Mass (1779).
